Sphaerisporangium flaviroseum sp. nov. and Sphaerisporangium album sp. nov., isolated from forest soil in China

Two Gram-positive, aerobic actinomycete strains YIM 48771T and YIM 48782T were isolated from virgin forest soil samples collected in Hunan Province, China. The two strains belong to the genus Sphaerisporangium according to a polyphasic taxonomic approach. Based on the phylogenetic analysis, similarities of 16S rRNA gene sequence of YIM 48771T with four valid published species, Sphaerisporangium viridialbum, Sphaerisporangium cinnabarinum, Sphaerisporangium melleum, Sphaerisporangium rubeum and YIM 48782T were respectively 97.3%, 97.4%, 97.2%, 96.3% and 97.1%, and those of YIM 48782T with the four type species were 96.9%, 97.6%, 96.9% and 97.1%. Results from DNA-DNA hybridization of the two strains with the four species revealed that DNA homologies of them were below 64.1%. MK-9 of the two strains was 28.1% and 29.0%, but those of the four type species are small amount. Contents of fatty acid C15:0, iso-C15:0 and C17:0 of the two strains were below 10%, and no C17:1{omega}8c, but S. viridialbum and S. rubeum are more than 10%. C16:0 of YIM 48771T was 15.02%, but the four species and YIM 48782T are below 10.3%. The two strains contented diphosphatidylglycerol, phosphatidylinositol mannoside, phosphatidylethanolamine and phosphoglycolipids, which are identical with the genus. Besides, strain YIM 48771T contented phosphatidylmethylethanolamine and phosphatidylinositol, and strain YIM 48782T phosphatidylinositol. G+C of YIM 48771T was 67.1%, and below those of the four species and YIM 48782T. So we propose two novel species, Sphaerisporangium flavoroseum YIM 48771T (=DSM 45170T =KCTC 19393T) and Sphaerisporangium album YIM 48782T (=DSM 45172T =CCTCC AA 208026T).
