Emended description of the genus Phyllobacterium and description of four novel species associated with plant roots: Phyllobacterium bourgognense sp. nov., Phyllobacterium ifriqiyense sp. nov., Phyllobacterium leguminum sp. nov. and Phyllobacterium brassicacearum sp. nov., by S. Mantelin, M. Fischer-Le Saux, F. Zakhia, G. Béna, S. Bonneau, H. Jeder, P. de Lajudie and J.-C. Cleyet-Marel

International Journal of Systematic and Evolutionary Microbiology vol. 56, part 4, pp. 827 - 839

Composition of media used

YM medium (l-1):
1 g yeast extract; 0.5 g sodium glutamate; 10 g D-mannitol; 0.5 g KH2PO4; 0.1 g MgSO4.7H2O; 1 ml 0.4 % (w/v) FeCl3; 1 ml 5 % (w/v) NaCl; 1 ml 4 % (w/v) CaCl2; pH 6.8.

Recommended medium for growth and maintenance of Ochrobactrum anthropi (l-1):
1 g beef extract; 2 g yeast extract; 5 g peptone; 5 g NaCl; pH 7.4.

Recommended medium for growth and maintenance of Chelatobacter heintzii (l-1):
1 g beef extract; 2 g yeast extract; 5 g peptone; 0.45 g KH2PO4; 2.39 g Na2HPO4.12H2O; 5 g NaCl; pH 6.8.

Recommended medium for growth and maintenance of Rhodobacter sphaeroides (l-1):
1 g yeast extract; 1 g disodium succinate; 5 ml 0.1 % (w/v) ferric citrate solution; 0.5 g KH2PO4; 0.4 g MgSO4.7H2O; 0.4 g NaCl; 0.4 g NH4Cl; 50 mg CaCl2.2H2O; 1 ml trace element solution (see below); 0.5 ml ethanol; pH 6.8.

Trace element solution (l-1):
0.1 g ZnSO4.7H2O; 30 mg MnCl2.4H2O; 0.3 g H3BO3; 0.2 g CoCl2.6H2O; 10 mg CuCl2.2H2O; 20 mg NiCl2.6H2O; 30 mg Na2MoO4.2H2O.

Distribute 40 ml medium into 50 ml screw-capped bottles. Flush each bottle for 1 to 2 min with nitrogen gas and then close immediately with rubber septa and screw-caps. Autoclave (120 °C, 20 min). Sterile syringes are used to inoculate and remove the samples. Incubate in light using a tungsten lamp.
