Supplementary zymogram

Zymogram of inorganic pyrophosphatase (PPase) activity from protein extracts of the serovar standard strains of ureaplasmas from humans after electrophoretic separation on a vertical slab polyacrylamide gel. The assay (Davis et al., 1987) indentified the site of phosphatase release after sodium pyrophosphate was supplied as substrate. Lanes 1–14 are cell-free extracts (15 micrograms protein) of the respective serovars; the PPase of cluster A strains (T960T biovar) had a smaller molecular weight than those from cluster B strains (parvo biovar). The lane designated 'M' contained 30 micrograms concentrated culture-medium components (15 micrograms protein) as a negative control, whilst the lane designated 'Pc' contained yeast PPase as a positive control (0.15 U).
